基于网络药理学研究银杏蜜环口服溶液对心脑血管保护的作用机制

摘要:

目的 采用网络药理学的思路和方法,研究银杏蜜环口服溶液发挥心脑血管保护作用的有效成
分、作用靶点、信号通路等,阐释其作用机制。方法 采用TCMSP数据库、SwissADME数据库筛选银杏蜜环口
服溶液的有效成分,在Swiss Target Prediction数据库和CTD数据库进行靶点预测和收集;通过GeneCards数据库
分别检索冠心病(CHD)、心绞痛(AP)和缺血性脑血管疾病(ICVD)相关靶点,将药物预测靶点和疾病
靶点取交集,再将3个疾病交集靶点对比取交集获得脑心同治靶点,利用Cytoscape软件构建“成分-靶点-疾
病”网络。运用String数据库,对交集靶点进行蛋白-蛋白相互作用(PPI)网络模型构建,找出核心靶点。通
过R软件对脑心同治交集靶点进行GO和KEGG富集分析。结果 银杏蜜环口服溶液筛选出24个活性成分、71个
成分-疾病交集靶点,可能作用的机制与PI3K-Akt信号通路等多个信号通路有关。结论 本研究探讨了银杏蜜
环口服溶液脑心同治的有效成分和作用机制,体现了中药多成分、多靶点、多途径的作用特点。

Abstract:

Objective To study the effective components, targets and signaling pathways of Yinxing
Mihuan Oral Solution for cardiovascular and cerebrovascular protection by applying ideas and methods of network
pharmacology, and explain the mechanism of its action. Methods The effective components of Yinxing Mihuan
Oral Solution were screened by using TCMSP database and SwissADME database, and targets were predicted and
collected in Swiss Target Prediction database and CTD database. The targets related to coronary heart disease (CHD),
angina pectoris (AP) and ischemic cerebrovascular disease (ICVD) were retrieved from GeneCards database. The
drug predictive targets and disease targets were intersected, and intersection targets of CHD, AP and ICVD were
compared and intersected to obtain brain-heart resynchronization therapy. A component-target-disease network
was established by using Cytoscape software. The network model of protein-protein interaction (PPI) on intersection
targets was established by using String database to find out core target. The intersection targets of brain-heart
resynchronization therapy were given GO and KEGG enrichment analyses by using R software. Results There
were 24 effective components and 71 component-disease intersection targets screened from Yinxing Mihuan Oral
Solution, and the possible mechanism of its action was related to multiple signaling pathways including PI3KAkt
signaling pathway. Conclusion The effective components and mechanism of action of Yinxing Mihuan Oral
Solution for cardiovascular and cerebrovascular protection are investigated, and the study reflects the characteristics
of multi-component, multi-target and multi-channel actions of traditional Chinese medicinals.
